      利用SSR分子标记技术,对扇贝“渤海红”、墨西哥湾扇贝及其杂交子代3个群体共90个个体的遗传多样性及遗传背景进行分析,结果发现:8个SSR位点共扩增出67个等位基因,各位点等位基因数范围为4-14个,平均等位基因数为8.5个,扇贝“渤海红”、墨西哥湾扇贝及其杂交子代群体的平均有效等位基因数(Ne)分别为:2.3947、2.048、1.8316;多态信息含量(PIC)分别为0.462、0.4373、0.358;平均期望杂合度分别为0.5045、0.2016、0.2458;3个群体的遗传分化指数(Fst)、基因流(Nm)、固定系数(Fis)分别为0.1398、1.5387、0.3698;杂交子代与亲本墨西哥湾扇贝遗传相似度和遗传距离分别0.7563 、0.2793,杂交子代与亲本扇贝“渤海红”为0.888、0.1188;研究结果表明:3个群体均具有中等的遗传多样性,扇贝“渤海红”遗传多样性最高,杂交子代与两亲本的遗传差异不对等,杂交子代的遗传结构更偏向亲本扇贝“渤海红”。
英文摘要:
      The hybridization between the “Bohai Red”and Argopecten irradians concentricus had been conducted and hybrids exhibit heterosis in survival rate, growth rate and high temperature tolerance, demonstrating substantial potential to replace A. irradians concentricus (Say).To better use of the hybrids, we need to understand the genetic structure of the hybrids and parents.Microsatellites is a widely used method to assess the genetic diversity in farmed aquatic species and construct QTL due to its characteristics such as the abundant polymorphism,the rich information, the co-dominance and conservation.In this research,the genetic diversity of 90 individuals in 3 populations of Argopecten irradians concentricus, “Bohai Red” and their hybrids were analyzed by 8 pairs of microsatellite primers. A total of 67 alleles were amplified and 4-14 alleles at each locus were generated, with a mean allele number of 8.5. The number of effect alleles (Ne), expected heterozygosity (He), and polymorphicinformation content were all highest in “Bohai Red” (2.3947, 0.504 and 0.462, respectively). The genetic differentiation index (Fst), gene flow (Nm) and fixed coefficient (Fis) was 0.1398, 1.5387 and 0.3698, respectively. The genetic distance between the hybrids and the “Bohai Red” is the smallest (0.1188), and the genetic similarity is the largest (0.888) which suggesting genetic structure of hybrids is close to “Bohai Red”.These findings will not only provide references for genetic evaluation of “Bohai Red” and Argopecten irradians concentricus ,but aslo for breeding of new varieties.
