为探究DNA甲基化在三疣梭子蟹副溶血弧菌感染中的表观遗传调控机制,对三疣梭子蟹PtRab2A进行了基因结构、相似性、系统进化、组织表达以及血淋巴组织在副溶血弧菌感染过程中不同时间点的基因表达水平和DNA甲基化模式进行了研究。结果表明:该基因全长为1824bp,其中包括15bp的5′ UTR,822bp的ORF区域和987bp的3′ UTR,在PtRab2A的C端发现了两个保守的半胱氨酸残基,预测分子式为C1322H2084N382O399S12,分子量为30098.13 kDa,理论等电点(PI)为7.05,共编码274个氨基酸。PtRab2A的保守结构域与拟穴青蟹的保守结构域相似性最高(100%);实时荧光定量PCR分析显示,PtRab2A基因在三疣梭子蟹各个组织中均有表达,副溶血弧菌感染下该基因的表达水平在12 h和72 h时显著升高 (P<0.05);BS-PCR分析显示,副溶血弧菌感染72 h后该基因的DNA甲基化水平显著降低,与基因表达呈显著负相关性(P<0.05)。采用RNAi敲降PtRab2A后,其表达量显著下调,24 h时,敲降效率达到80%。干扰后注射副溶血弧菌后发现,实验组的死亡率在2 h时显著高于对照组,且死亡高峰期提前。因此,副溶血弧菌感染下,三疣梭子蟹可能通过DNA甲基化促进免疫基因的表达,从而提高其免疫力。研究结果丰富了三疣梭子蟹抵御病原入侵的分子调控机制。 |

In order to investigate the epigenetic regulatory mechanism of DNA methylation in Vibrio parahaemolyticus infection in Portunus trituberculatus, gene structure, similarity, phylogeny, tissue expression, and gene expression levels and DNA methylation patterns of hemolymph tissues at different time points during V. parahaemolyticus infection in P. trituberculatus PtRab2A were investigated. The results showed that the full length of the gene was 1824 bp, including a 5′ UTR of 15 bp, an ORF region of 822 bp and a 3′ UTR of 987 bp, two conserved cysteine residues were found at the C-terminal end of PtRab2A, with a predicted molecular formula of C1322H2084N382O399S12, a molecular weight of 30098.13 kDa and a theoretical The conserved structural domain of PtRab2A showed the highest similarity of 100% to the conserved structural domain of Scylla paramamosain.
