      综合运用肉眼观察、活体压片-醋酸洋红染色和组织切片-H.E.染色3种方法对大菱鲆Scophthalmus maximus L.幼鱼进行性别鉴定。结果表明,在所有采集的1 217尾全长8.3~21.5 cm、体重9.0~183.9 g的大菱鲆幼鱼样品中,体重>90 g的269尾幼鱼和体重10~90 g的790尾幼鱼,可用肉眼直接观察鉴定性别;体重10~90 g的另外154尾幼鱼,可采用活体压片方法鉴定性别;体重<10 g的4尾幼鱼,性腺难以剥离,则需组织切片的方法鉴定性别。对活体压片性别鉴定结果进行组织切片抽样验证表明,体重>10 g的幼鱼,均可以采用活体压片方法准确地鉴定出性别,其准确率可以达到100%。本研究验证了采用活体压片 醋酸洋红染色技术在大菱鲆幼鱼生理性别鉴定中具有很高的准确性,它不仅有利于全雌育种中对幼鱼快速进行性别鉴定,而且可以应用于商品苗性别的快速鉴定。

      In this study three combined methods including microscopic examination, aceto-carmine staining of squash mounting and haematoxylin-eosin staining of histology section of the gonads were adopted to determine the phenotypic sex of turbot Scophthalmus maximus L. young fish. Among the 1 217 individuals of turbot young fish (total length 8.3~21.5 cm, wet body weight 9.0~183.9 g), all of the 269 individuals weighting more than 90 g and 790 individuals weighting between 10~90 g could be unambiguously sexed by macroscopic examination of their gonads, whereas the remaining of 154 individuals weighting between 10~90 g and 4 individuals weighting less than 10 g were identified using squash mount preparation and histology section, respectively. The accuracy of the squash mount observation of young fish weighting more than 10 g is up to 100 percent, which was validated by classic histology section. With the validated efficiency and accuracy in the sexing of turbot young fish, the squash mount technique holds out the prospect of considerable improvement in the development of the production technique of all female turbot juvenile.
