棘头梅童鱼染色体核型分析

摘要:
以棘头梅童鱼(Collichthys lucidus)为实验材料,经胸腔注射植物血球凝集素(PHA)和秋水仙素,取其头肾组织,进行低渗、固定,采用空气干燥法制片,吉姆萨染液染色,观察棘头梅童鱼染色体核型和特征。结果显示,棘头梅童鱼染色体数目为2n=48,占所观察分裂相的83.3%;核型公式为2n=48t,染色体臂数NF为48,染色体相对长度范围为(5.694±0.514)~(3.039±0.161),属于鲈形目原始核型,且符合典型的高位类鱼类核型特征。研究中未发现与性别相关的异型染色体、次缢痕及随体等。本研究为棘头梅童鱼的细胞遗传学研究提供了基础资料,并为其种质鉴定提供参考数据。

Karyotype Analysis of Collichthys lucidus

Abstract:
In this study, the metaphase chromosomes of Collichthys lucidus were obtained from kidney tissue by the method of PHA and colchicine injection-air drying method. The experimental fishes were taken from Ningde coast of China. Because of their strong stress, we adjusted the drug treatment time (PHA: 6 h; Colchicine: 2 h, 5 h). In order to identify whether there were heteromorphic chromosomes, we separated the female from the male. The results showed that there were 48 chromosomes in C. lucidus and its karyotype formula was 2n=48t, NF=48. The relative length of chromosome was in the range of (5.694±0.514)~(3.039±0.161), which suggested that C. lucidus was in accordance with the original karyotype of perciformes and the higher group of fish evolutionary taxonomy. Sex chromosome, satellite chromosome and secondary constrictions were not found in C. lucidus. This study provides basic data for cytogenetics and germplasm identification for C. lucidus.
